    XSLT Implementation in Relational Database Environment

    XML is widely used format for storing all kinds of data and XSLT standard represents a standardized way, how to transform a XML document to a di erent structure. Many XSLT implementation has been introduced, but the most of them uses an in-memory representation of the transformed XML document. The implementation done in this thesis uses the relational database engine to store processed document and takes advantages of SQL to evaluate XPath expressions used by XSLT. First, importing source XML document into the generic relational mapping is described. For processing XPath expressions, the XPath to SQL convertor is introduced. Lastly, the processing of XSLT stylesheets by relational database engine is shown.XML je rozšiřitlný formát, který je využíván k ukládání všech druhů dat, a XSLT představuje standardizovaný jazyk pro transformaci XML dat a jejich struktury. Dnes existuje mnoho implementací XSLT, ale většina z nich udržuje zdrojové XML dokumenty ve strukturách přímo v paměti. Implementace představená v této práci používá pro udržování těchto dokumentů relační databázi a využívá jazyk SQL k vyhodnocování všech XPath dotazů, které se používají v XSLT. Nejdříve je představeno nahrávání zdrojových XML dokumentů do generického relačního mapování.     This thesis is focused on the topic of the computer program as an employee work and its specifics. The aim of this thesis is mainly describing and summarizing the Czech legislation of the employee work in the Copyright act if such work is the computer program including its history. The thesis also contains the author's remarks with respect to the relevant contractual practice. In the introductory passage of this thesis there is an explanation of the basic term "computer program" and its usage in the Czech legislation together with the explanation of the basic terms regarding the copyright such as "author work and its origin", "authorship", "content of copyright", "disposition of the content of copyright" etc. Then there is a short excursion to the databases and their copyright protection. The following parts of this thesis focus on the crucial author's work which is the computer program wherein is included the excursion to the history of the copyright protection of the computer program as the author's work followed by the excursion to the international and the European regulation of the computer program as the author's work. Then the relevant provision of the Czech regulation regarding the employee work is discussed in detail which is followed by a chapter containing some of the author's practical..

    I na začátku 21. století má textová příkazová řádka v operačních systémech stále pevné místo. Obvyklé je to u systémů založených na Unixu, kde její schopnosti často zcela překonávají možnosti grafického uživatelského rozhraní. Platí to zejména pro jednu její často používanou schopnost - pracovat efektivně s filtračními nástroji. Používání příkazové řádky, řetězů a filtrů ovšem není jednoduché a klade velké mnemotechnické nároky na uživatele. Tato práce se zabývá myšlenkou, jestli je možné tuto podmnožinu funkčnosti příkazové řádky uživateli usnadnit pomocí interaktivního grafického rozhraní. Smyslem této práce je na něj definovat uživatelské požadavky, zjistit problémy spojené s jeho tvorbou a pokusit se je vyřešit. Cílem je pak využít těchto závěrů a vytvořit funkční prototyp programu, nazvaný SPsim (Shell Pipe Simulator).At the beginning of 21st. century, the text command line still has its own place in operating systems. This is often the case with those based on Unix, where its CLI capabilities often completely surpass abilities of graphical user interface. This is especially true for one of its commonly used functionalities - to work effectively by using the filter commands. Using of the command line, pipes and filters, is, though, not an easy task, and is putting a a lot of mnenonic requirements on user. This thesis is concerned about whether it is possible for such subset of functionalities to be simplified for user by use of graphical representation. Goal of this thesis is to define requirements for such environment, find out possible obstacles connected with achieving this task, and try to solve them. Final product of this work is to utilize such findings and creation of a working prototype of such program, called SPsim (Shell Pipe Simulator)

    Field Use of Protective Bacteriophages against Pectinolytic Bacteria of Potato

    The pectinolytic Dickeya solani bacterium is an important pathogen found in potatoes. We conducted laboratory and field experiments mimicking severe and mild Dickeya spp. infection and investigated the application of a mixture of two lytic bacteriophages before and after bacterial infection to protect the plants. Application of the phage solution to tuber disks and wounded tubers did not completely eliminate the infection but reduced the development of soft rot symptoms by 59.5–91.4%, depending on the phage concentration. In the field trial, plants treated with bacteriophages after severe Dickeya infection had 5–33% greater leaf cover and 4–16% greater tuber yield compared to untreated plants. When simulating a mild infection, leaf cover was 11–42% greater, and tuber yield was 25–31% greater compared to untreated plants. We conclude that the phage mixture has the potential to protect potatoes ecologically from D. solani

    Background and aimGene defects contribute to the aetiology of intrahepatic cholestasis. We aimed to explore the outcome of whole-exome sequencing (WES) in a cohort of 51 patients with this diagnosis.Patients and methodsBoth paediatric (n = 33) and adult (n = 18) patients with cholestatic liver disease of unknown aetiology were eligible. WES was used for reassessment of 34 patients (23 children) without diagnostic genotypes in ABCB11, ATP8B1, ABCB4 or JAG1 demonstrable by previous Sanger sequencing, and for primary assessment of additional 17 patients (10 children). Nasopharyngeal swab mRNA was analysed to address variant pathogenicity in two families.ResultsWES revealed biallelic variation in 3 ciliopathy genes (PKHD1, TMEM67 and IFT172) in 4 clinically unrelated index subjects (3 children and 1 adult), heterozygosity for a known variant in PPOX in one adult index subject, and homozygosity for an unreported splice-site variation in F11R in one child. Whereas phenotypes of the index patients with mutated PKHD1, TMEM67, and PPOX corresponded with those elsewhere reported, how F11R variation underlies liver disease remains unclear. Two unrelated patients harboured different novel biallelic variants in IFT172, a gene implicated in short-rib thoracic dysplasia 10 and Bardet-Biedl syndrome 20. One patient, a homozygote for IFT172 rs780205001 c.167A>C p.(Lys56Thr) born to first cousins, had liver disease, interpreted on biopsy aged 4y as glycogen storage disease, followed by adult-onset nephronophthisis at 25y. The other, a compound heterozygote for novel frameshift variant IFT172 NM_015662.3 c.2070del p.(Met690Ilefs*11) and 2 syntenic missense variants IFT172 rs776310391 c.157T>A p.(Phe53Ile) and rs746462745 c.164C>G p.(Thr55Ser), had a severe 8mo cholestatic episode in early infancy, with persisting hyperbilirubinemia and fibrosis on imaging studies at 17y. No patient had skeletal malformations.ConclusionOur findings suggest association of IFT172 variants with non-syndromic cholestatic liver disease.</div
